In clinical trials the response of a subject to one of two treatments may depend upon a covariate. Estimates of the difference in the expected responses as a function of the covariate with minimum mean squared error depend upon the model used and the allocation of the treatments to subjects. For nearly linear models, minimax estimates are obtained and an optimum allocation procedure is described.
